Effects of different smoke extracts on retrobulbar fibroblasts
measured by real time cell electronic sensing technique

Introduction: Cigarette smoke exposure is a known risk factor for thyroid associated orbitopathy (TAO), with serious effect on disease se-
verity and response to treatment. Nowadays, alternative forms of smoking, such as heated tobacco products (HTPs) or e-cigarettes (ECIGs),
are becoming more common alongside traditional cigarettes (CCs).
Aim: Analysis of primary fibroblast cell culture from tissue removed from TAO and healthy orbits with the addition of smoke extracts from
different tobacco products.
Methods: Primary cell cultures were obtained from tissue collected during orbital surgery. Cells were examined for 72 hours with continuous
measurement in DMEM medium by adding tobacco smoke extracts (SE), which were prepared according to previous publications. The
metabolic activity of the cells was assessed by MTT assay and hyaluronan (HA) production by ELISA at 24, 48, 60 and 72 h after the
addition of extracts.
Results: Untreated TAO cells had the lowest cell index (CI) value (CI max =3.3). CC treated TAO CI values peaked at 5.38. HTP and
ECIG treated TAO cells behaved similarly and reached higher values compared to CC treated cells. ECIG treated TAO cells had CI max =6.33
and HTP treated TAO cells had CI max =6.25. TAO cells treated with CC, HTP and ECIG all showed significant differences (p<0.0001).
The CI values show a time-dependent, dynamic change in cellular activity, with ECIG causing the largest CI change in TAO fibroblasts.
There was no significant difference in metabolic activity in any of the SE treated cell lines compared to untreated cells. The basal metabolic
activity of untreated GO and NON-GO cells did not differ. No difference was found in HA production between treated and untreated cells.
Conclusion: The orbital fibroblasts responded differently to CC, HTP and ECIG smoke extracts and cells from TAO samples were more
sensitive to tobacco smoke treatments than normal fibroblasts. The current model may be useful for designing further in vitro researches to
investigate the pathogenesis of TAO, and the negative effects of smoking.

dasági kihívások miatt a területen folyó intenzív kutatás-fejlesztés számos fontos előrelépést jelentő újdonságot eredménye-
zett mind a diagnosztika, mind a terápia területén. A kurzus előadója áttekinti a diagnosztikus szemlélet változásait, a vál-
tozások technikai követéseként fejlesztett új eszközök nyújtotta lehetőségeket, továbbá bepillantást nyújt saját kutatásaik
jövőbeni gyógyszerfejlesztési és klinikai felhasználásra is alkalmasnak ítélt eredményeiről.

Dry eye disease: novelties from the bench to bedside

Dry eye disease has become an emerging problem of ophthalmology in the industrialized countries. Socio-economic difficulties caused by
the disease positioned this morbidity in the front line of ophthalmic research and development resulting in numerous new outcomes both in
the diagnostic and therapeutic fields. The course will review the latest innovations of diagnosis and the new approaches of dry eye disease
enabled by the new diagnostic devices. The author will also give an overview of her own latest scientific results which might be in the scope
of pharmacological or clinical utilization in the future.
